    The Contribution Of Subunit Interfaces To Receptor Activation In Ligand Gated Ion Channels

    This project seeks to provide insights into new mechanisms that could be used to enhance or inhibit neuronal signalling. The family of pentameric neurotransmitter receptors that are key components in the process of neuronal signalling and are the target of this study. It will investigate the molecular motions that occur when the receptor shifts from the resting state to the activated state in the presence of neurotransmitter.     Using MiR-200 To Find New Therapeutic Targets For Neuroblastoma

    Neuroblastoma is one of the most common cancers in children. We have found that a genetic regulator, called microRNA, can limit the ability of neuroblastoma cells to invade surrounding tissues and metastasise. We aim use the microRNAs to find new therapeutic targets that may work in combination with existing treatments, reducing the short term toxicity and long term deleterious effects of current treatments.

    Identifying Mitochondrial Genome Variants Associated With Familial Migraine Susceptibility

    New therapeutic targets for migraine are desperately needed. Although studies have identified some migraine genes there remains considerable underlying genetic variation to be characterised. This study aims to identify functional variants in the mitochondrial genome that contribute to migraine susceptibility, utilising the isolated Norfolk Island population. Outcomes will determine the significance of the variants identified, potentially leading to new diagnostics.

    Identifying Novel Gene Mutations For Molecular Diagnosis Of Familial Hemiplegic Migraine

    This proposal aims to identify novel FHM genes by undertaking an NGS screen of the whole exome of 209 FHM patient samples. We will test the pathological relevance of detected novel mutations by functional analysis in human cell models and using patient-specific stem cell techniques. Using whole genome NGS technology to identify novel mutations will assist in the design and development of a comprehensive NGS approach to diagnose and differentiate this severe neurological disorder.

    Melanotransferrin: A “Missing Link” And A Novel Pharmacological Target For Treatment

    Despite >30 years of research, the precise function of the protein, melanotransferrin (MTf), is unknown. However, we have breakthrough evidence that MTf stimulates WNT signalling as a major driver in cancer progression. We will investigate this hypothesis, which will underpin new cancer therapies. Indeed, we designed a new class of drugs that target the WNT pathway via up-regulating the WNT inhibitor, NDRG1. This drug (DpC) inhibits MTf expression to block tumour cell growth and metastasis.

    Neisseria gonorrhoeae, the pathogen that causes gonorrhoea, has developed resistance to multiple classes of antibiotics. The last remaining class of antibiotics that can successfully be used for general treatment of gonorrhoea is extended spectrum cephalosporins (ESCs). However, we are now starting to see Neisseria gonorrhoeae isolates with early signs of ESC resistance. This research will use molecular technology and mathematical modelling to investigate Neisseria gonorrhoeae antimicrobial resi ....     Genetic Variants Underlying X-linked Familial Migraine

    VRE is a serious hospital superbug that has been increasing in many major hospitals around Australia, while at the same time MRSA (Golden Staph) infections have been decreasing. This project will find out why VRE is increasing by examining what happens to patients at a major Australian hospital from their time of admission to the onset of infection with VRE. At the end of the project we will have the first real understanding of how VRE is transmitted so we can develop effective infection control ....
