    A Study Of Artemisinin Combination Therapy Given At Delivery To Prevent Postpartum Malaria And To Young Infants To Treat Uncomplicated Malaria

    Summary
    The proposed studies will investigate the preventive value of a course of combination antimalarial treatment at delivery in pregnant women in malarial areas. The transfer of this treatment into breast milk and to the suckling infant will be investigated since this may protect the infant against malaria but also cause drug-related side-effects. These data will be used, with a study of combination treatment in infants with malaria, to optimise dose regimens in this vulnerable group.

    Previous NHMRC-sponsored research from the applicants has demonstrated that one of the strongest risk factors for subsequent development of asthma is having chest infections during infancy that are so severe that they trigger symptoms of fever and wheeze. It is not known what predisposes susceptible infants to these severe infections, and this project will attempt to define the mechanisms of susceptibility.

    Vaccination during pregnancy can offer protection against severe respiratory disease for infants in the first six months of life. For this reason, influenza and pertussis vaccines are routinely recommended during each pregnancy. Unfortunately, little is known about the ‘real world’ effect of both vaccines. We plan to conduct the largest and most comprehensive study to date to evaluate all vaccines routinely recommended in pregnancy in Australia.

    Characterisation Of Eurl, A Novel Gene Implicated In The Etiology Of Abnormal Brain Development And Intellectual Disability

    Intellectual disability affects around one per cent of Australians, and can arise from genetic abnormalities during fetal life, such as through abnormal regulation of gene expression. We have identified a novel gene, known as eurl, which controls brain assembly as well as the ability of neurons to form functional connections within the brain. We will investigate how this novel gene controls brain development, and characterise eurl as a potential therapeutic target for learning and memory.

    The timely differentiation of neural stem cells is critical during development, and the unrestrained proliferation of neural stem cells in the adult can lead to deadly brain cancers such as glioma. At present our understanding of the key molecules that regulate neural stem cell behaviour during these processes remains limited. In this proposal we will investigate the molecular determinants underpinning neural stem cell biology, both within the developing brain, and within glioma.

    Infection with group A streptococcus (GAS) is a major cause of morbidity and mortality worldwide, including in the Aboriginal population of Australia. Concerted efforts for vaccine development have been hampered by the absence of a suitable animal model. To address this critical knowledge gap we propose to develop a controlled human infection model of GAS infection. This model will provide a direct pathway for the future appraisal of novel GAS vaccines.

    Understanding The Causes Of Childhood Congenital Anomalies Of The Kidney And Urinary Tract

    Summary
    Congenital anomalies of the kidney and urinary tract (CAKUT) is a common cause of renal failure in children. The majority of patients with CAKUT do not know the underlying cause of their renal anomalies. In this proposal we will characterise the developmental events that are perturbed in three mouse models of CAKUT and identify the causal gene responsible in each mouse model. We will translate this information to the clinic by screening patients with CAKUT for mutations in these newly identified ....     REACH: Randomised Trial Of EArly Rehabilitation In Congenital Hemiplegia

    Infants with asymmetric brain lesions are at high risk of congenital hemiplegia. This study compares modified CIMT to an equal dose of bimanual training in 150 infants recruited at 3-6 months. Both therapies will be parent-delivered supported by experienced clinicians. Outcomes include use of the impaired hand in bimanual tasks, cognitive and motor development at 12 and 24 months c.a. with measures of neural structure and functional connectivity at 24 months.
