Summary
There are more than 7 million overweight or obese adults in Australia. With conventional methods of weight loss, only 5% of these people will be able to achieve permanent weight reduction of any significance to health outcomes. This project aims to develop more effective and permanent methods of weight management through comparing effectiveness of continuous versus intermittent dieting; and determining what metabolic, hormonal and behavioural factors predict weight regain.

Brown fat, unlike ordinary fat generates heat by burning calories to maintain body temperature. Originally thought to disappear after birth, recent research including our own have shown that brown fat exists in adult humans in varying amounts. Our aim is to find ways of turning on people�s brown fat allowing them to lose weight by burning more calories.
Achieving More Effective Weight Loss With Intermittent Energy Restriction

Summary
Part of the difficulty in achieving lasting weight loss is that the body responds to dieting with compensatory changes in appetite, metabolic rate & blood hormone levels. We have shown in men that alternating phases of energy restriction with periods of greater food intake markedly improves weight loss by overcoming this “famine reaction”. This study will examine if the intermittent diet also produces more effective weight loss in women, and whether it is applicable to the wider community.
I am a physiologist focused on the role of dietary protein on excess weight gain. In the proposed study, I will examine energy balance in lean and overweight individuals in response to a 4-day covertly manipulated 10, 15 or 25% protein diet. We will measure effects on hunger using subjective ratings and hormone levels and the affects on energy expenditure including resting metabolic rate, the energy it takes to digest food and physical activity levels.
Chronic Kidney Diseases (CKDs) present serious morbidity and mortality in our society. Kidney scarring is the final manifestation of many types of kidney diseases. Recent evidence showed that a reduced ability to generate energy during injuries leads to kidney scarring. My study is looking at how the specific changes in kidney energy production can lead to kidney scarring. The findings of my study can have potential to prevent kidney scarring and change the course of kidney diseases.

Obesity is an increasing problem in the community and is associated with an increased incidence of high blood pressure, high cholesterol and triglycerides, diabetes, heart disease and cancer. Although conventional dietary advice for weight loss is a low fat high carbohydrate and high fibre diet there is a very strong interest in the media and the general population in alternative diets that feature reduced carbohydrate with the remainder of the diet either protein or a mixture of protein and fat ....Obesity is an increasing problem in the community and is associated with an increased incidence of high blood pressure, high cholesterol and triglycerides, diabetes, heart disease and cancer. Although conventional dietary advice for weight loss is a low fat high carbohydrate and high fibre diet there is a very strong interest in the media and the general population in alternative diets that feature reduced carbohydrate with the remainder of the diet either protein or a mixture of protein and fat. Unfortunately there is very little published data to guide the decisions of health professionals in this area and there is a strong demand for such information. We plan to perform 2 large weight loss studies which focus on high protein diets with one exchanging protein for carbohydrate and keeping fat constant and the second exchanging protein for fat keeping carbohydrate constant. We will assess whether the high protein diets blunt the decrease in metabolic rate that occurs with weight loss, and whether it spares some of the inevitable loss of muscle mass. Anorexia nervosa (AN) is an eating disorder characterized by behavioral alterations, restrictive eating leading to extremely low body weight. AN is also associated with a paradoxical hyperactivity representing a failure of the brain to adjust activity to energy status. Here we aim to investigate the role of the Neuropeptide Y (NPY), a known regulator of homeostatic control and activity in the development of hyperactivity in AN, thus help to identify NPY-targeted therapeutics for AN treatment.

Summary
A large body of evidence for the health benefits and life-extending properties of dietary restriction exists. Recent findings suggest that periods of fasting can have beneficial effects, even without an overall reduction in caloric intake. This proposal will compare periodic fasting with and without weight loss, versus daily caloric restriction on metabolic health outcomes in humans and examine mechanisms that may contribute to this effects.
Many heart diseases are associated with impairment of energetics of the heart. Improving the heart's energetics can lead to improved survival and long-term outcomes. Perhexiline is a heart medication that works by improving the way the heart uses energy. Although effective, it is associated with long-term toxicities. Better understanding of this medication may lead to less adverse effects and also provide a basis for further investigation of drug development in the future.
