Summary
Facility for multidimensional fractionation of complex biological mixtures. Acquisition of multidimensional fractionation equipment will allow researchers to separate proteins from complex mixtures, and to compare whole protein profiles of multiple samples. This will permit correlation of specific protein changes associated with infection or disease, a major focus of post-genomic programs of research. The equipment will also provide identification of the key differentiating proteins using mini ....Facility for multidimensional fractionation of complex biological mixtures. Acquisition of multidimensional fractionation equipment will allow researchers to separate proteins from complex mixtures, and to compare whole protein profiles of multiple samples. This will permit correlation of specific protein changes associated with infection or disease, a major focus of post-genomic programs of research. The equipment will also provide identification of the key differentiating proteins using minimal material. Numerous world-class projects and researchers will be able to move more rapidly and reliably from crude cell extracts to identifiable markers, and maintain their competitive positions the recognition of key targets in drug design, disease diagnosis and vaccine development.Read moreRead less
High resolution single particle analysis of biological macromolecules. One of the great challenges of cell biology is to increase the rate of atomic resolution structure determination, particularly of membrane proteins and macromolecular assemblies. One of the great challenges of cell biology is to increase the rate of atomic resolution structure determination, particularly of membrane proteins and macromolecular assemblies. The current rate-limiting step is high quality crystal production. Our goal is to prove that protein structures can be determined to atomic resolution by single-particle analysis. 3D structures will be produced by computationally aligning high-resolution electron microscope images of individual, randomly oriented molecules. The importance of this project is highlighted by the fact over 120,000 protein sequences are already databased, a number set to increase rapidly as new genome sequencing projects are completed.

Peptidic spider toxins: a novel paradigm for control of insect pests. Many insects are serious pests of Australian crops, livestock, and pets. Australian farmers spend about $300 million per annum on insecticides and acaricides, while Australian consumers spend more than $100 million annually on insecticides for use around the home and garden, and on pets. Viruses disseminated by insects are also responsible for diseases such as dengue, Japanese encephalitis, and Ross River fever. Unfortunately, many of these insect pests have developed resistance to chemical insecticides. The aim of this research program is to develop a new generation of environmentally-friendly natural products and insect-resistant crops that can be used to control insect pests on farms and around the home and garden.Read moreRead less
Safeguarding Australia against invasive arthropod pests. An increasingly serious public health issue for Australia is the emergence of infectious diseases disseminated by arthropods such as ticks and mosquitoes. Arthropod-borne viruses are already the major human pathogens in Australia, and they disproportionately affect Aboriginal communities. The aim of this research is to develop environmentally-sustainable methods for controlling arthropods that destroy crops or disseminate human and animal disease. These insecticides will not only provide benefits within Australian territories, but will be useful to our defence forces when operating in overseas locations where arthropod pests are a problem (e.g., malarial regions of Iraq).Read moreRead less

Novel methods of manufacturing biodegradable polymer scaffolds around which new tissue can be grown within the human body will be developed. Surfactant - polymer assemblies will be used to produce highly porous scaffolds of tunable pore size and connectivity, shape and strength. The results will create a new avenue for systematic investigations into the effects of scaffold structure on tissue growth. This research will lead to the development of reliable, well-controlled manufacturing techniques for tissue engineering scaffolds, revolutionising current scaffold manufacturing practices. It will enhance existing collaborations between the University of Melbourne and the Bernard O'Brien Institute of Microsurgery.Read moreRead less
Structural studies of plant disease resistance proteins. Plant cells have evolved a gene-for-gene disease resistance mechanism, involving an interaction of a plant-derived receptor with a specific pathogen-derived molecule. Currently, plant breeders are restricted to the resistance genes available in particular crop species or sexually compatible relatives. Plant cells have evolved a gene-for-gene disease resistance mechanism, involving an interaction of a plant-derived receptor with a specific pathogen-derived molecule. Currently, plant breeders are restricted to the resistance genes available in particular crop species or sexually compatible relatives. In the last few years, several plant disease resistance genes have been identified, providing a foundation for studying the molecular basis of the resistance process. We propose to obtain three-dimensional structural information on representative R proteins and their ligand complexes. This will form the basis for modifying existing resistance genes to confer resistance to new diseases, resulting in large economic benefits.Read moreRead less

Drug discovery is reliant on the application of basic research and advanced technologies to obtain solutions to human suffering caused by diseases. This application seeks funding to support the installation of state-of-the-art mass spectrometry instrumentation at Griffith University and the University of Melbourne. The requested instrumentation will enhance high quality collaborative research programs in drug discovery and eradicate significant deficiencies in research infrastructure at both institutions. Access to state-of-the-art mass spectrometry instrumentation will markedly improve our international competitiveness across a broad range of biological and chemical science disciplines.Read moreRead less

Management of crop diseases involves the integrated use of resistant cultivars and the application of chemical pesticides. Many diseases, however, including rust, continue to pose an economically significant threat to agricultural productivity in Australia. The research outlined in this proposal aims to understand the mechanisms, at a molecular and structural level, that enable resistant plants to detect and respond to pathogen attack. The outcomes of this currently unavailable fundamental understanding will enable new, durable and more effective resistance genes to be engineered. Therefore, the work has significant economic and environmental implications for agricultural crop plant productivity in this country.Read moreRead less
