    Structural Studies On The Immune Effector Perforin: Developing Mechanism-based Inhibitors

    Perforin is an essential weapon deployed by the human immune cells in order to destroy virally infected or cancerous cells. Despite this key role, unwanted or excessive perforin function can result in disease and can severely impact on successful treatment of leukaemia through bone marrow transplantation. This application aims to understand the molecular details of perforin function, and to apply this knowledge to develop perforin inhibitors.

    The Claustrum Enigma: Unlocking The Role Of The Last Unknown Area Of The Primate Brain

Interest in this brain structure has recently been revived by findings showing damage to the claustrum in several major diseases, and by the suggestion that the claustrum may have a role in consciousness. Here we propose a series of experiments to test the novel hypothesis that the claustrum is part of the default mode network, a group of brain areas that act together when we aren't thinking about anything in parti ....     ASPREE is the largest clinical trial ever conducted in Australia and will determine whether daily low dose aspirin prevents disease in healthy older people. The study was well-funded initially but will require additional support to complete the vital final stage of data collection and analysis. This will enable the study to answer important questions about the benefits and risks of aspirin in this age group and its effect on disability free survival.

    A Randomised Controlled Trial To Improve Depression In Family Carers Through A Physical Activity Intervention

    This RCT of a physical activity intervention for carers and care recipients aims to determine whether physical activity (designed for carer and care recipient to do together) can reduce depression in older carers. The intervention includes a physiotherapist assessment, Otago and Otago-Plus exercise programs in carer/care recipient’s home over six-months. An economic evaluation will be undertaken. If successful, the intervention could be broadly applied within the carer support service system.

    An International Comparison Of Systems Of Care, Risk Stratification And Outcomes In TIA And Minor Stroke

    Transient ischaemic attacks (TIAs: self-limited brief episodes of stroke symptoms) are high-risk events with an approximately 15% risk of stroke within 90-days. Current practice calls for specialist treatment within 1-7 days. But this recommendation (based on 20 year old overseas evidence) may not apply in contemporary Australia. We will conduct a study of patients with TIA that will document their interactions with Australian health systems and inform contemporary Australian practice.

    A Randomised Clinical Trial Of Physical Activity To Delay The Progression Of Cerebrovascular Pathology

    The aim of this randomised clinical trial is to establish whether a 24 months physical activity (PA) program for older adults with memory problems can delay the progression of damage to the blood vessel system in the brain, measured on an MRI scan. It also will investigate whether cognition, mood, quality of life, functional level, fitness and biological markers will improve with the intervention. If successful this PA program could become part of clinical care for adults at risk of dementia.

    Loss of independence is common after stroke, and may lead to reduced quality of life and admission to nursing home care. We will investigate if an increased amount of rehabilitation following stroke leads to improved mobility. Two ways of delivering more intense rehabilitation will be compared with usual care to find out which leads to improved physical mobility, and how they compare economically. This will allow health service providers to optimise services and will benefit people with stroke.
