Examining New Treatments And Developing New Treatment Biomarkers For Youth With Severe Depression

Antidepressant medications and psychotherapy have been the mainstays of depression treatment in young people, but given their modest effectiveness, there is a pressing need for new treatment strategies. During this fellowship I aim to examine better treatments for depression, and develop better predictors about who is likely to benefit from them.
A Selective Prevention Trial Using Novel Pharmacotherapies In An Older Age Cohort At Risk For Depression

The study will examine whether omega-3 fatty acids or antidepressants prevent the onset of depression in a group of older community participants who have previously been identified as being "at risk" of depression. The study will incorporate sophisticated brain scanning methods, as well as tests of brain functioning to determine acute brain changes, reduction in cognitive decline and prevention of depression over a one-year period.
Mood and anxiety disorders represent a tremendous cause of disability and morbidity. This research will use cutting-edge brain imaging technologies to understand why these disorders typically emerge in young people and whether brain imaging can reliably predict the effectiveness of different treatments for individual patients.

Depression and bipolar disorder are two of Australia’s leading causes of disease burden, with 3-4 million Australians affected during their lifetime. However, despite being such a serious health burden, there are major shortcomings to current understanding and management. This research program aims to further our knowledge of the causes and presentations of the mood disorders, and to improve and “tailor” treatment for the many Australians who suffer from these highly disabling illnesses.

This project investigates the role of epigenetic processes (i.e., functional changes to the genome affecting gene expression) in mediating the long-term effects of childhood trauma in schizophrenia and bipolar-I disorder. We specifically aim to distinguish the effects of trauma-related epigenetic effects on brain structure and function, and on the immune and stress-response systems, and the potential moderation of these effects by structural genetic variants associated with psychosis risk.
A Randomised Controlled Trial Of Low-dose Ketamine In Youth With Severe Depression And Elevated Suicide Risk

Recent research has shown that a single injection of low-dose ketamine has powerful, though short-lived, antidepressant effects. Effective treatments are urgently needed for young people with severe depression. This will be the first controlled study to test whether repeated doses of ketamine, given over 4 weeks, is effective for young patients.

Borderline personality disorder (BPD) is a severe mental disorder with adverse long-term outcomes, including suicide. BPD is characterised by vulnerability to stressful life events and catastrophic responses to stress. This study of youth early in the course of BPD examines the baseline biological characteristics of the stress response system, how these characteristics might be influenced by treatment and how this relates to treatment outcome. The findings will inform early intervention for BPD.
