    Developmental Origins Of Adult Cardiovascular Disease: Vascular Health In The Raine Cohort

    Summary
    The Raine study is a unique long term experiment that has collected extensive pre-birth and childhood data in ~3000 young Australians, who are now 27 years old. We plan to measure the artery health of 1200 of these volunteers and to determine what factors, both before and after birth, influence the presence of early atherosclerosis in humans. This study will guide strategies aimed at early prevention of heart attacks and stroke in humans, by defining the major risk factors.

    M-PreM Study: Reproductive Factors, From Menarche To Pre-menopause, And The Risk Of Cardiometabolic And Respiratory Conditions Before Menopause

    This study will investigate links between reproductive factors and body size of premenopausal women with their risk of major chronic conditions, such as cardiovascular disease, diabetes, and asthma. It builds on two decades of survey data from Australia’s flagship study of women’s health with a new biomedical assessment. Findings will guide the use of indicators of women’s reproductive health as a trigger for early and targeted approaches for chronic disease prevention.

    Clinical Review Of A Cohort Aged 22-33 Years Conceived Using Assisted Reproductive Technologies

    In a recent study, using telephone-interviews, we compared the health and wellbeing of 547 singleton young adults born following assisted reproductive technologies (ART), with 549 matched controls. Reviewing their health when they are 22-33 years is possible because of their ongoing interest. We have a protocol in place to measure their cardiac and respiratory function and other aspects of growth and development. Our findings will fill a major knowledge gap about the longer term safety of ART.

    The Structural Basis For Glutamate Transporter Function

    Glutamate transporters are vacuum cleaners in the brain that suck the neurotransmitter glutamate into cells. When the glutamate vacuum breaks down or becomes blocked, glutamate levels outside cells increase, leading to cell death in the brain. This process underlies the damage in many brain diseases including Alzheimer’s disease and stroke. The aim of this project is to understand the mechanism of the glutamate vacuum cleaner so we can develop therapeutics to fix it when it breaks down.

    Signaling Pathways To Enhance Potency Of AMPK-targeting Drugs

    Sedentary lifestyles and consumption of high energy foods has led to epidemics of obesity-related metabolic diseases that place enormous financial and medical burden on the Australian economy. An attractive drug target to treat these diseases is AMP-activated protein kinase (AMPK) which functions as both a cellular fuel gauge and co-ordinator of whole-body metabolism. Our goal is to improve AMPK drug potency by identifying novel processes that sensitize AMPK to drugs.

    The GOOD NIGHT Project: A Sleep Intervention To Prevent Depression And Improve Cardiac Health In Adolescence

    There is a pressing need for effective approaches to preventing depression during adolescence. The primary aim of the project is to assess whether a brief sleep intervention can prevent depression in vulnerable adolescents. Given that sleep interventions also encourage daytime activity, which when combined with improved sleep has potential benefits for cardiovascular health, the secondary aim is to test whether the sleep intervention improves early indicators of cardiovascular risk.

    Do Exposures Before Conception Influence The Risk Of Asthma In Offspring?

    Asthma and poor lung function are major causes of public health issues. Emerging evidence suggests adverse exposures even before the conception of a child may cause these conditions. The proposed project is part of an international study across generations to identify these factors. These original findings will be of great importance both nationally and internationally.

    Cardiovascular disease (heart attack and stroke) are leading causes of death and illness in adults in Australia. The changes in blood vessels that lead to these conditions begin before birth.     Improving The Neonatal Transition In Infants With A Congenital Diaphragmatic Hernia

    Summary
    Congenital diaphragmatic hernia is a common congenital abnormality and occurs when the diaphragm fails to separate the abdominal and thoracic compartments before birth. This prevents the lung from growing properly and so at birth, the lung is unable to take over the role of gas exchange without considerable assistance. As a result, these infants are at high risk of death or significant disability and this application is focused on improving care and reducing morbidity in these infants.

    Clinical Trial Of A Suprachoroidal Visual Prosthesis For The Profoundly Vision Impaired

    Summary
    For 15 years we have been designing a bionic eye. We have made a device called the Phoenix99 and shown in short term animal tests that it is both safe to implant but also that it potentially performs better than any other device in the world. We are requesting funds to complete longer term animal testing of the device and then commence a small human clinical trial to demonstrate the benefits of the technology – specifically that it is able to help blind people navigate without assistance.
