This proposal builds on my lab's recent discovery that E-cadherin interacts with and regulates the Arp2/3 actin nucleator complex, a central determinant of actin assembly in cells. We will explore key implications of this finding, concentrating on defining the molecular mechanisms that regulate Arp2/3 and actin assembly in cadherin-based adhesion. Our work combines molecular characterization of regulatory mechanisms and proteomic searches for new regulators, with tests of the higher-order function of this novel process in cell adhesion and recognition.Read moreRead less

Dissecting a hematopietic transcription factor complex. The development of mature active cells is a highly complex and coordinated process that is controlled largely by groups of interacting regulatory proteins. We are trying to understand, at a very detailed level, how a specific group of these proteins interact to regulate both normal blood cell development and the onset of childhood leukemias. Using this information we will try to develop reagents that can be used to inhibit these interaction ....Dissecting a hematopietic transcription factor complex. The Shape of Plants; Discovering factors that control morphology by organizing the cytoskeleton. Understanding how plants generate the huge diversity of shapes seen in nature is both a scientific challenge and a biotechnological opportunity. Microtubules dominate cell architecture, providing dynamic, yet rigid, frameworks for defining or changing growth polarity. Understanding how plants generate the huge diversity of shapes seen in nature is both a scientific challenge and a biotechnological opportunity. Microtubules dominate cell architecture, providing dynamic, yet rigid, frameworks for defining or changing growth polarity. We recently discovered and cloned MOR1, a gene that is essential for organizing microtubules and controlling morphogenesis. This places us in a strong position to resolve a long-standing mystery: how are microtubules organized? We intend to define MOR1's structural attributes, identify its interacting proteins and innovate an ambitious screen for additional genes that have related functions. This project should stimulate new ideas and applications.Read moreRead less
Tissue tension homeostasis by junctional mechanosensing. This project examines how tissues use mechanical tension to preserve their integrity. This comes from the recent appreciation that cells pull on the connections between each other to generate tension. Further, molecular mechanisms exist for cells to sense changes in this tension and then to enlist the appropriate responses to restore tension. The project aims to test how local changes in tension are detected and corrected, when tissue inte ....Tissue tension homeostasis by junctional mechanosensing. Further, molecular mechanisms exist for cells to sense changes in this tension and then to enlist the appropriate responses to restore tension. The project aims to test how local changes in tension are detected and corrected, when tissue integrity is compromised by very different causes. The project endeavours to establish a new conceptual paradigm for understanding tissue homeostasis, based on cell biology and biomechanics, with implications for developmental biology and tissue engineering.Read moreRead less
Versatile elastin based hybrid hydrogels for chondrocyte transplantation and repair. Cartilage repair is often limited after significant trauma, sports injury and disease. This project will generate a new family of hybrid biomaterials constructed by precisely blending natural and synthetic components. These novel biomaterials will establish the foundation for manufactured prefabrication and in situ injection to promote healing.

The present project focuses on the proteins making cellulose and how they organised into functional complexes that are able to make cellulose. The knowledge it provides, together with that from other projects, will move us towards the situation where we can manipulate the rate at which cellulose is produced and change its detailed properties. This opens the way to industry producing fibres with more desirable properties and producing novel cellulose-based materials tailored to specific applications.Read moreRead less

Reaction intermediates along hydrolytic pathways and molecular determinants of substrate specificity of barley B-glucan exo- and endohydrolases will be defined using crystallographic and kinetic analyses. These enzymes are of central importance in cell wall metabolism during development of higher plants, and in plant-pathogen interactions. Realization of the project objectives will not only provide fundamental information on catalytic mechanisms, but will also provide opportunities to manipulate enzyme specificity. Further, site-directed mutagenesis of the enzymes will be used to generate glycosynthases, which will be evaluated for their ability to synthesise novel oligosaccharide and polysaccharide products, some of which might show immunomodulating activity.Read moreRead less
