This study focuses on key endocrine pathways involved in the remodelling of the breast stromal cells into a reactive stromal environment which is more permissive for tumour growth. We have identified key pathways involved in the regulation of estrogen biosynthesis and fibrosis in tumour associated stroma. These studies will lead to the development of novel breast cancer therapies.

We all talk, eat and drink, but it’s not until we lose these skills that we recognise their importance. Communication & swallowing disorders following from progressive brain disorders such as dementia and ataxia result in daily disadvantage through poor educational and employment outcomes & social isolation. These deficits make the evaluation, long term monitoring and treatment a critical element in disease management.
This project aims to improve the acute treatment of children with burn injuries through developing more effective and novel treatments and understanding the conditions required for burn injuries to occur.

Summary
Frontotemporal dementia (FTD) and motor neuron disease (MND) are fatal and incurable neurodegenerative diseases. Although dissimilar in symptoms, most cases of both FTD and MND are characterised at autopsy by abnormal accumulations of the TDP-43 protein in neurons. In this fellowship, I will characterise the biochemical changes caused by TDP-43 malfunction that occur in both FTD and MND and test potential new therapeutics in mice, to identify new ways to treat these devastating diseases.
Characterisation And Therapeutic Targeting Of Molecular Pathways That Promote Breast Cancer Metastasis To Bone

Summary
Breast cancer that has spread to bone cannot be cured. Using the most clinically relevant model of breast cancer available we have identified that tumour cells growing in bone need to suppress immune elimination (by suppressing the Type I interferons) and invade through the bone tissue (by activation of cysteine cathepsins). Studying the functional role of these pathways will provide novel insight into the mechanisms of breast cancer spread to bone that can be augmented therapeutically.
Development Of Endogenous Granulocyte Colony Stimulating Factor (G-CSF) Antagonism As A New Therapeutic Approach To Inflammatory Disease

Summary
Neutrophils play a pivotal role in inflammatory diseases including rheumatoid arthritis (RA). G-CSF is a growth factor that is important to neutrophil survival and function. We have shown that in the absence of G-CSF the incidence and severity of experimental autoimmune arthritis are reduced. We will investigate the mechanisms by which this occurs as well as studying the effects of G-CSF blockade on function and survival of human neutrophils from healthy donors and RA patients.

Breast cancers are made up of different types of cancer cells, and not all cells contribute equally. A subset of cancer cells may be uniquely capable of driving tumor growth, rebuilding fatal tumors after therapy and establishing new tumors at distant sites. Identifying and exploiting the pathways that regulate the activity and survival of these cells will lead to better modes of treatment, and move towards a relapse-free future for breast cancer patients.
Fighting Mental Disorders: Using Mental Imagery To Disarm Symptoms And Enhance Treatment.

This proposal will conduct the first pre-clinical and clinical research utilising mental imagery to disarm symptoms and enhance treatment of multiple mental disorders by controlling the strength of mental imagery. This fellowship represents the first attempt to alleviate symptoms by developing a cognitive-neuroscience method to control the strength of traumatic imagery and to show a causative means of enhancing imagery strength to boost the success rate of cognitive therapy involving imagery.
