    Genome-wide Association Studies To Identify Major Genetic Determinants Of 5 Blinding Eye Diseases Using Pooled DNA

    Summary
    This project aims to find important genes for 5 diseases that can lead to blindness. We will use a cost-effective approach where samples from a large number of individuals with a given disorder are pooled (mixed together) and then compared on gene chips covering the whole genome to a pool of people who do not have the disease. Differences identified between the groups will point to genes causing that disease. We will identify any major genes for the 5 diseases being studied.

    The Functional Organisation And Signals Of Motion Sensitive Neurons In The Middle-temporal Area Of Visual Cortex

    Some nerve cells in the cerebral cortex are very sensitive to visual motion. These neurons direct eye movements and provide motion perception, but the "neuronal code" they carry is poorly understood. We will address this basic question in experimental studies of the primate visual system. This project will help us understand visual performance; poor motion vision is an early indicator of many neurological disorders and this knowledge can help develop methods for their detection and diagnosis.

    A possum in a tree is cute to see on a bushwalk, but scary to hear when we are trying to sleep. This illustrates that how we perceive a “target” is affected by “modulators” that are close in space or time to the target, or by the task at hand. Deficits in contextual modulation are apparent in many neurological conditions. This project will investigate the neural circuitry that mediates spatial, temporal and task-related contextual modulation.

    Safety And Efficacy Of A Surgically Implanted Suprachoroidal Retinal Prosthesis (Bionic Eye)

    A bionic eye is a electronic device which can stimulate the remaining visual pathway in a person who is blind, to restore some basic vision. Our team have previously shown that our novel bionic eye device can be safely implanted in a patient, and can give improvements in vision when tested in a laboratory environment. The next stage of the research is to provide patients with a more advanced device, which will contain more electrodes and also be able to be taken home.

    There are an estimated 60 million glaucoma sufferers worldwide. Glaucoma is associated with elevated pressure within the eye but the mechanisms by which this leads to vision loss are not clear. We propose to test a new hypothesis that it is tissue stress that is responsible for the ganglion cell death in glaucoma. We propose to measure key parameters of the cellular environment in an animal model of raised intraocular pressure and determine if elevated intraocular pressure induced changes in the ....     The project aims to use the principle of "predictive coding" - a theory that promises to be a unified theory of the brain - to understand how the visual cortex makes predictions about future events, at cellular level.     Corneal opacities and scarring due to microbial and parasitic infections are a major cause of blindness globally. Novel studies in our lab have shown that topical application of bacterial/viral DNA alone to the cornea can cause previously unrecognised inflammation in the retina. Understanding the mechanisms of this retinal inflammation and how to block it may help in the design of novel treatments for a number of blinding conditions.

    Giant cell arteritis (GCA) is the most common form of vasculitis in people over 50 years of age. If untreated it can cause catastrophic complications including blindness, though this can be prevented if treated early. Although there is clear evidence for a role of genetic factors in GCA, these have been little studied. We have established an Australian-led International consortium, with clinical, basic science and statistical expertise to thoroughly investigate this devastating disease.

    Migraine is a common neurological condition affecting approximately 15% of adults. Therapies are most effective if used early, yet many people are unable to predict their migraines or to recognize early signs. In addition to headache, key symptoms include abnormal visual and auditory experience. We propose that aspects of vision and hearing will vary systematically according to the current brain status. Our translational goal is to develop tests that allow individuals to better manage migraine.

    Blinding eye diseases with genetic contributing factors affecting the retina or light-sensing part of the eye, damage the vision of millions of people worldwide. The vision impairment is irreversible, untreatable, and worsens with age. In this project, we will use technological breakthroughs in genome correction and sequencing, in combination with stem cell and mouse model applications, to develop pathways to treatment of these currently incurable diseases.
