The overall aim is to improve treatments and outcomes for people with osteoporosis. This will be achieved by better predicting those who are likely to fracture and subsequently those who do well post fracture from those who do poorly. Following an osteoporotic fracture there is an increased risk of re- fracture and of premature death. This research will define those risk factors for fracture, re-fracture and early death in a large group of men and women followed for over 20 years.

Despite decades of research, the underlying genetic mechanisms for why it occurs are not well understood. The study will involve substantial Aboriginal leadership and consultation and will be a model for the conduct of genetic studies in Aboriginal populations.

Summary
Understanding the genetics of Alzheimer’s disease (AD) is one of the best ways of improving our knowledge of the underlying mechanisms of disease development. There is evidence that genetic factors account for up to 80% of the risk for AD. Recent advances in genetics still explain <50% of this risk. This proposal will help two major Australian studies to partner with a large European consortium in trying to understand the “missing” heritability, using existing and novel genetic data.
Rapidly giving intravenous fluid to prevent or treat shock (fluid resuscitation) is one of the commonest treatments given to critically ill patients. Current guidelines recommend crystalloid solutions but it is unknown whether any particular crystalloid is better than others. This trial will determine whether the use of one of two crystalloid fluids, saline or PlasmaLyte, reduces the risk of organ injuries, such as kidney failure, and improves patients chances of surviving critically illness.

Its use in Australia has been severely limited by poor outcomes compared to the rest of the world. The burden associated with heroin dependence is undeniable. But little is known about the natural history and long-term course of heroin dependence; knowledge that is critical for informing the development of new treatment interventions, health care planning and service delivery. We propose to extend our study of 615 Australians with heroin dependence, recruited in 2001-2002, to 18-20 years follow-up to answer critical questions about the long-term impact of this condition.
