Mechanisms of manchette function. This project aims to define the function of the manchette, a poorly understood microtubule-based structure present in haploid male germ cells. This project aims to define key mechanisms underpinning manchette development and movement, and to generate a detailed picture of the dynamics of germ cell development using imaging technologies and unique animal models. Such knowledge should improve the understanding of how male fertility is achieved, the origin of infertility and how species-specific differences in sperm form are achieved. Such insights may ultimately lead to improved agricultural efficiencies and job creation.Read moreRead less
Improving the efficiency of bovine oocyte maturation in vitro. For the dairy and beef industries, the hundreds of eggs (oocytes) in a high value cow's ovary that fail to produce a pregnancy are a wasted genetic resource. A key technology to unlocking this resource is in vitro maturation of oocytes, but the process is inefficient. One possible cause is that the current approaches to maturing oocytes in vitro do not adequately mimic the natural process in vivo. One possible cause is that the current approaches to maturing oocytes in vitro do not adequately mimic the natural process in vivo. We will design new systems to mature cow oocytes in vitro by altering the chemical composition of maturation medium, thus improving the efficiency of laboratory embryo production and related technologies.Read moreRead less

This research proposal offers a pioneering opportunity to develop treatments that overcome the negative impact of stress on reproduction. Specifically, knowledge generated in this project will be vital in the development of strategic defences against the impact of stress on reproduction. This project fundamentally addresses Research Priority 2: Promoting and maintaining good health. Given that suppression of reproduction by stress occurs in all mammalian species including humans, domestic animals and wildlife, being able to prevent or overcome stress-induced reproductive dysfunction will generate significant health, social, economic and ecological benefits. A novel microtubule severing protein involved in male germ cell biology. The project aims to better understand the cellular and biochemical mechanisms underlying a key component of male fertility. Microtubules are a fundamental component of all cells. A mechanism that is increasingly recognised as essential for microtubules regulation is severing. It has been discovered that an uncharacterised microtubule severing protein, KATNAL2, has a key role in male germ cell development. A mechanism that is increasingly recognised as essential for microtubules regulation is severing. It has been discovered that an uncharacterised microtubule severing protein, KATNAL2, has a key role in male germ cell development. This project aims to define the mechanisms underlying KATNAL2 function in the male germ line. It is expected that these data will generate a comprehensive picture of KATNAL2 function and provide foundation data of relevance across multiple species and tissues. In the longer term, it may also reveal a rational strategy for fertility enhancement or suppression.Read moreRead less
In vitro prediction tests of fertility of frozen semen of sheep and cattle. Variation in fertility after artificial insemination (AI) of frozen semen is high and leads to economic loss. Routine assessment of the quality of frozen semen relies on crude, simple tests of motility and morphology of sperm. More sophisticated sperm ultrastructural and function tests have recently been developed, but it is not known how these tests relate to fertility in the field. Routine assessment of the quality of frozen semen relies on crude, simple tests of motility and morphology of sperm. More sophisticated sperm ultrastructural and function tests have recently been developed, but it is not known how these tests relate to fertility in the field. In this project we will test a range of in vitro methods of assessment of motility and morphology of sperm and compare them with fertility of frozen semen samples after AI, leading to more objective methods of semen analysis.Read moreRead less

To date there is no available information on the underlying biochemical and genetic mechanisms that control gender and fertility in Thenus spp. Understanding how gender and fertility are controlled in this lobster species will allow development of technologies to exploit desired commercial traits such as reproductive sterility (for genetic protection). Such technologies will facilitate rapid uptake of commercial culture of this high-value species in Australia and establishment of international markets. Read moreRead less
Sex preselection of stallion spermatozoa. This study will develop novel techniques that will maintain and optimise the fertility of sex-sorted stallion semen before and after processing through a flow cytometer. Ultimately, spermatozoa obtained from ejaculation or from the testes sperm storage site (epididymes) at the time of castration will be sorted into X- (female) or Y- (male) populations and used to inseminate mares and thus, enable horse breeders to predetermine the sex of their foal at th ....Sex preselection of stallion spermatozoa. The introduction of sex-sorted semen into the Australian horse industry (worth some $6.2 billion) will ensure that Australia leads the world in equine reproductive technology.Read moreRead less

The recruitment of primordial follicles into the growth phase is central to female reproductive function, however the control of this process to date, has been poorly understood due to inadequate technologies. Our team has recently developed novel recruitment models and a new and innovative method of isolating primordial follicles which will enable us to identify the role of c-kit and selected TGF beta family members in recruitment. This work will provide cornerstone scientific knowledge about the control of female reproduction and provide the impetus for the development of more effective contraception and superovulation strategies in mammals.Read moreRead less

The dairy industry will not deliver sustainable higher productivity unless the reproductive performance of dairy cows is significantly improved. This expected outcome will be achieved via the identification of epigenetic and protein biomarkers of improved fertility that will be used to develop superior breeding stock. Caruncular and intercaruncular tissues and blood have been obtained from both fertile (New Zealand Holstein-Friesian) and sub-fertile (North American Holstein-Friesian) cows within pasture-based dairying systems that include seasonal calving and subclinical uterine inflammation (associated with poor reproductive performance). Determinations will be conducted using deep parallel (next generation) sequencing and mass spectrometry.Read moreRead less
Interactions between cells and extracellular matrix in the epithelial-mesenchymal transition of the ovarian follicular stratified epithelium. The lining of many mammalian organs and cavities contain cells that can transform into different cells to bring about organ development or repair but if it goes horribly wrong the cells become metastatic cancers. This project examines the key features of this process especially the roles of matrix that develops around the cells in this process.
