HEN1 is a regulator of piRNA metabolism, transcriptional regulation and mammalian male fertility. This project is to define the biochemistry of a previously uncharacterized protein in male fertility using a unique mouse model and innovative DNA and protein technologies. This project will define a novel, and essential, pathway for male fertility and may ultimately have relevance to the maintenance of health or improving fertility.
Novel mechanisms integrating the central and autonomic nervous system. This project aims to define molecular mechanisms controlling the exquisite connectivity of neurons in different parts of the body. The ability of higher-vertebrates to respond to different environmental conditions is essential for life, evolution, health, reproduction and growth, and is reliant on the autonomic nervous system. This project aims to define molecular mechanisms controlling the exquisite connectivity of neurons in different parts of the body. The ability of higher-vertebrates to respond to different environmental conditions is essential for life, evolution, health, reproduction and growth, and is reliant on the autonomic nervous system. However, how the autonomic nervous system is integrated with the central nervous system to control holistic physiological responses is largely unknown. By deciphering how neural networks are formed this project aims to provide broad biological insight to wiring of the entire nervous system which is likely to have significant implications for the formation of synthetic neural networks and for regeneration.Read moreRead less

This project aims to develop statistical methods that can explore genotype–environment interaction at the genomic level using genome-wide single nucleotide polymorphisms or sequence data. It plans to estimate how the effects of genetic variants change with changing environmental conditions and how overall genetic variance changes due to changing effects in specific gene regions. It plans to deliver statistical models and methods and an efficient algorithm implemented in software, which would broadly benefit the field of complex trait genetics. Methods to estimate genotype–environment interaction effects at the genomic level would help elucidate complex biological systems, including human genetic response to changing environmental factors and the potential adaptation of animals to changing environmental conditions.Read moreRead less
Regressive evolution of eyes in subterranean water beetles of arid-zone Australia: A comparative phylogenetic approach. Reduction or total loss of non-functional characters are common evolutionary phenomena, but little is known of the genetic basis of this regressive evolution. This project will use a phylogenetic framework to investigate the forces responsible for evolution of eyelessness in subterranean water beetles, by studying molecular genetic variation in three key eye developmental genes. The water beetles are explicitly suitable to address the questions due to their numerous independently evolved eyeless species and relatively old age of divergence from surface relatives. The research will provide a major new perspective on regressive evolution and the relationship between gene structure and function.Read moreRead less
Genetic control of spermatogenesis: defining the role of SOX3 in spermatogonial progenitor cells. The transcription factor (TF) SOX3 is a key regulator of neural stem/progenitor cells. Recently, this project has also shown that SOX3 is active in sperm progenitors (spermatogonia) and is required for spermatogenesis. Using our Sox3 KO mouse model and extensive expertise in spermatogonial cell culture, ChIP-seq technology and bioinformatics, this project will investigate crucial aspects of SOX3 fun ....Genetic control of spermatogenesis: defining the role of SOX3 in spermatogonial progenitor cells. Recently, this project has also shown that SOX3 is active in sperm progenitors (spermatogonia) and is required for spermatogenesis. Using our Sox3 KO mouse model and extensive expertise in spermatogonial cell culture, ChIP-seq technology and bioinformatics, this project will investigate crucial aspects of SOX3 function in the testes including stem versus progenitor cell activity and genome-wide target gene regulation. These studies will uncover the molecular and cellular mechanism by which SOX3 controls spermatogenesis and provide unique insight into how a single TF controls context-dependent differentiation in sperm versus brain progenitor cells.Read moreRead less

Provision of dedicated instruments for contemporary and ancient/fragmentary DNA analyses will provide numerous opportunities for innovative research solutions in basic biology, archaeological, agricultural, biomedical, forensic and environmental sciences. No similar combination of facilities currently exists in the Australian region severely curtailing and jeopardising the quality of current and proposed research programs. The facilities will underlie innovative approaches to research in National Research Priorities 1 and 4 - An Environmentally Sustainable Australia and Safeguarding AustraliaRead moreRead less
The evolutionary origin, cellular response and genetic impact of insecticide resistance mutations in agricultural pests. Diamondback moth is a global pest of canola and Brassica vegetables, and populations can rapidly evolve resistance to insecticides. The project will use a combination of genome sequencing, fieldwork and gene expression analysis to identify mutations causing resistance to Bt insecticidal toxins and assess the threat of resistance evolving in Australia.

This project aims to develop whole genome approaches that can improve the estimation and prediction power by using information from the dynamic genetic architecture of complex traits (i.e. the changes of genetic characteristics and effects when varying effective population size and genetic backgrounds). The project intends to deliver advanced statistical models, efficient algorithms and design by combining data from close relatives, population samples or from different populations (e.g. multi-ethnicities or multi-breeds). The expected outcome is to better understand the dynamic architecture of complex traits and develop methods with improved power, precision and accuracy in genomic analyses.Read moreRead less

Sustaining crop yield and maintaining food security is a significant worldwide concern. This project aims to strengthen insect pest control strategies and improve bio-insecticide use in agriculture through better understanding of the mode of action of Bacillus thuringiensis (Bt) insecticides. It aims to improve their efficacy and evaluate opportunities to develop bio-insecticides based on plant saponins. This will assist in determining the risk of insecticide resistant moths migrating to Australia, and within our borders. This project aims to provide opportunities to improve transgenic Bt-crops and Bt-sprays, provide commercial development of new bio-insecticides, and develop optimal control strategies for major Australian migratory pests.Read moreRead less
