This investigation examines variations in the genes that are important determinants of human skin pigmentation and are likely to be associated with skin cancer risk. Our research program will form the basis of future diagnostics based on major genes that determine a persons skin type. Current skin cancer prevention strategies rely predominantly on broad spectrum campaigns that are aimed at increasing the general community awareness of the damaging effects of UV radiation. A better understanding of the genetic basis of UV-sensitive skin types will greatly enhance the targeting of such skin cancer-prevention campaigns, provide an understanding of changes that occur in skin pathology, and the mechanisms of sun induced tanning.Read moreRead less

Laboratory-grown human skin equivalent constructs, given social and legislative imperatives, will be critical for advances in novel treatment protocol definitions for wound repair, dermatogical screening of pharmacueticals and fundamental studies of skin diseases.
In silico studies undertaken in this project will make a significant contribution to the effectiveness of the application of human skin constructs, by delivering new and deeper insights into the interplay between dependent processes that regulate the behaviour of skin, in vivo or ex vivo. The models and the researchers associated with this project will drive innovative studies in medical science over the next decade.Read moreRead less
Transdermal penetration of corticosteroids in the dog. Topical application of corticosteroids enhances drug concentration and effectiveness in the treatment of skin diseases. Topical application of corticosteroids enhances drug concentration and effectiveness in the treatment of skin diseases. Most topical corticosteroid preparations have been developed for human use and are poorly efficacious or promote a high incidence of adverse effects in dogs. This project will characterize corticosteroid penetration through canine skin to permit the development of suitable topical formulations to more effectively control skin diseases in the dog. Skin diseases are a significant problem in veterinary science and this project will not only provide an effective therapeutic option, but also reduce animal (and client) distress when suffering skin disease and/or adverse effects from traditional therapy.Read moreRead less

VitroGroR is a growth factor complex which enhances cell growth and migration, and hence has great potential for treating wounds. Tissue Therapies, which holds the rights to commercialization of VitroGroR, is seeking to develop methods of delivering VitroGroR in its active form to the wound environment. Two solutions to this problem will be developed in this project; a bioactive bandage containing a novel combination of microspheres and a hydrogel matrix, and secondly an in-situ polymerisable matrix for treatment of deep wounds. The growth factor complex will be protected from aggressive proteases through encapsulation within microspheres, and the use of MMP-inhibiting comonomers.Read moreRead less
Studies in cancer control. As life expectancy in Australia (and throughout the world) continues to rise, so will the burden of cancer escalate. Treating cancer after diagnosis is costly, and in many instances, unsuccessful. As life expectancy in Australia (and throughout the world) continues to rise, so will the burden of cancer escalate. Treating cancer after diagnosis is costly, and in many instances, unsuccessful. Preventive strategies promise to reduce the future cancer burden, yet our knowledge in this arena is limited by the lack of credible research as to what works and what does not. This application addresses this gap directly by conducting research into the control of two cancers which exact a growing toll in Australia and elsewhere. The work seeks to identify and understand the causal pathways to cancer, and then use this information to devise evidence-based strategies for cancer control.Read moreRead less
Photoactive Semiconducting Biopolymers. The basic aims of this project are to elucidate, manipulate, and utilise the unique chemical and physical properties of a class of biopolymers called the melanins. These materials are the only known solid state semiconducting biopolymers, and are non-toxic, biocompatible, and biodegradable. Their use as active components in biomimetic soft electonic, optoelectronic or photovoltaic devices, has not hitherto been demonstrated. These materials are the only known solid state semiconducting biopolymers, and are non-toxic, biocompatible, and biodegradable. Their use as active components in biomimetic soft electonic, optoelectronic or photovoltaic devices, has not hitherto been demonstrated. It is anticipated that the key outcomes from the project will be a demonstration of biopolymer-based photoelectrochemical and solid-state p-i-n solar cells, and an improved understanding of the physics and chemistry of these important biological macromolecules.Read moreRead less

The outcomes of this project will be relevant to range of wounds, including burns, bed-sores, and diabetic and venous ulcers. Clearly, innovation in wound healing is urgently required as the treatment of wounds represents a significant challenge at all levels of our society, in terms of cost (physical, emotional and financial) to patients, the economy and to the wider community. Despite this obvious and overwhelming need, research in wound healing is relatively under-developed and has yet to adopt modern biotechnology and biomaterials approaches. This project is therefore directed at generating cost-effective frontier wound dressing technologies that accelerate wound repair.Read moreRead less

This is the first attempt to characterise the differences in human pigmentation using a combined genetic and cellular analysis of melanogenesis. We have the ability to culture the pigmenting cells of the human epidermis and hair follicles called melanocytes from individuals of defined genotype. This will allow us to correlate mutations in melanosomal proteins with functional defects within the cells in culture using live cell imaging, electron microscopy and biochemical analysis. This will provide a molecular basis to explain the pigmentary characteristics of individuals allowing prediction and diagnosis of their photosensitivity with important implications for skin cancer risk.Read moreRead less

Relationship between melanosome distribution and skin colour. This work seeks to examine how the colour of our skin is related to the distribution of melanosomes. This work may allow us to better understand how to develop novel therapies that may alter skin colour and provide protection from the sun (frontier technologies) but may also, in the longer term, lead to healthier approaches to skin management - especially for ageing skin.
