X-ray absorption spectroscopy (XAS) is an extremely important synchrotron radiation tool for determining the local structure around an X-ray absorbing atom. This has many applications in the study of materials, minerals, metal complexes, and metalloproteins and can often be used to obtain information that is not available by other techniques, because structural information can be obtained in the solid or solution state and in mixtures. The current proposal is aimed at introducing new technology into the Australian National Beamline Facility that will greatly improve the quality and quantity of experiments that can be performed and extend studies into dilute solutions and protein samples.Read moreRead less

This project has direct impact on pharmaceutical research: Biomacromolecular interactions are key points for pharmaceutical intervention and detailed structural knowledge of dynamic protein interactions can significantly accelerate drug development. Australia has invested in expensive instrumentation that can be used with new laboratory methods to obtain information on delicately balanced biomacromolecular interactions, and how they malfunction in disease. This project will provide a computational framework to increase the impact of this investment by integrating measurements from a range of novel technologies and developing understanding of changes in structure of large protein complexes in different functional states.Read moreRead less

By means of its high resolution and (MS)n capabilities, the FT-ICR-MS will provide key structural information on a wide range of synthetic and natural chemical substances, including sequence (e.g. peptides) and fragmentation patterns, while the MALDI-TOF instrument will be used primarily for high through-put proteomic analyses.Read moreRead less

This application seeks to exploit our established lead in the measurement of diffuse scattering from disordered materials and its analysis using large-scale computer simulations. Many industrially important materials such as ceramics, superconductors, catalysts, electro-optical materials and minerals owe their special properties to the disorder in their structure. This disorder causes diffuse X-ray scattering which can be probed using synchrotron X-rays and powerful computers to reveal details about the material's nanoscale structure. The new methodology being developed will enhance our detailed understanding of the relationships between structure and properties in materials and help promote the design of new materials. Access for Australian Researchers to Advanced Neutron Beam Techniques. Neutron scattering is one of the most powerful and important investigative tools in the study of materials. Australia has only a low-flux neutron source, HIFAR, which provides no cold or hot neutrons. This excludes large, important areas of science, such as functional films, polymers, self-assembly systems, biological materials, colloids and emulsions, and real-time in-situ studies.

This application aims to continue Australia's partnership with the world's most intense neutron source, ISIS in the UK, in order to sustain the considerable Australian scientific momentum which now relies on ISIS.
The outcomes will be new science that cannot be generated solely within Australia.

Designer Surfactants for Creation of Emulsion Properties. We propose new approaches to the design of surfactants for stabilising water in oil emulsions. X-ray and neutron scattering techniques, developed by us, allow the interfacial structure of the surfactant to be found and the intellectual "loop" closed in the design process. Preliminary screening will be done using the surface balance and bulk emulsion synthesis.Read moreRead less

The proposed research will lead, through a better understanding of polymorphism, to more efficient production of pharmaceuticals and will enhance the establishment and protection of patents. The work will have flow-on in other areas such as the manufacture of pigments, dyes and explosives. The project uses methodology for the elucidation of local structure and function at the atomic to nanoscale level in which Australia is a world leader. The project will further enhance our standing in this field and will provide excellent research training opportunities in areas particularly pertinent to future exploitation of the Australian Synchrotron and the new Research Reactor OPAL, which open in 2007.
