This investigation examines variations in the genes that are important determinants of human skin pigmentation and are likely to be associated with skin cancer risk. Our research program will form the basis of future diagnostics based on major genes that determine a persons skin type. Current skin cancer prevention strategies rely predominantly on broad spectrum campaigns that are aimed at increasing the general community awareness of the damaging effects of UV radiation. A better understanding of the genetic basis of UV-sensitive skin types will greatly enhance the targeting of such skin cancer-prevention campaigns, provide an understanding of changes that occur in skin pathology, and the mechanisms of sun induced tanning.Read moreRead less

Innovative agricultural solutions in Australia's future will be built on understanding and manipulating the expression of groups of genes to influence whole plant phenotypes providing more robust plants and high value plant products. Plant energy organelles are central components in plant metabolism, their coordination by processes such as dual-targeting has potential to modify germination characteristics, early seedling vigour, and stress tolerance. Studying energy organelles could generate valuable intellectual property to be applied within Australia's large plant-based industries and at the same time provide a rich intellectual environment for the training of research students and postdoctoral researchers.Read moreRead less

Identifying novel insecticides and their targets: probing Australian arachnid venoms. Insect pests destroy an estimated 2-3 billion dollars of crops in Australia. Insect pests also are responsible for the transmission of many new and re-emerging human, animal and plant diseases threatening health, wellbeing and prosperity. Current insecticides are severely limited by toxicity and/or insect resistance, and some are undergoing use cancellation overseas. Thus there is an urgent need to develop safe ....Identifying novel insecticides and their targets: probing Australian arachnid venoms. Thus there is an urgent need to develop safer and more specific insecticides that are effective against disease vectors and agricultural pest insects, as well as to identify new insecticide targets. This research has a multi-million dollar potential benefit to agricultural, health and pest control sectorsRead moreRead less
The regulation of signalling molecules in Saccharomyces Cerevisiae by inositol polyphosphate 5-phosphatases. Phosphoinositide signalling molecules regulate the actin cytoskeleton, secretion, vesicular trafficking and cell growth and death. Phosphoinositide signalling molecules regulate the actin cytoskeleton, secretion, vesicular trafficking and cell growth and death. We have identified, cloned and characterised a family of signal terminating enzymes called inositol polyphosphate 5-phosphatases (5-phosphatases) that regulate phosphoinositide signalling molecules. We have cloned and characterised four distinct 5-phosphatases in the yeast Saccharomyces Cerevisiae and demonstrated by both deletion and overexpression studies that these enzymes regulate the actin cytoskeleton, endocytosis and secretion. This research proposal aims to investigate the signalling complexes the 5-phosphatases form with specific actin binding and or regulatory proteins, investigate the complex interactions of phosphoinositide lipid phosphatases and the roles they play in regulating secretion from the endoplasmic reticulum and finally characterize a novel 5-phosphatase that we have recently identified. Collectively the outcome of these studies will provide novel information about the functionallly significant signalling pathways regulated by this important enzyme family.Read moreRead less

Non-classical steroid signalling through SF-1 responsive genes: a key mechanism in environmental endocrine disruption, cancer, and aging. Endocrine disruption by pervasive manmade chemicals, which mimic natural hormones, and are found in plastics, cosmetics, and fire retardants, is known to cause developmental defects in model organisms and wildlife, with substantial risk also to human health. This risk increases with increasing population density and dependence on water recycling. Current tests to assess such substances use oversimplified modes of hormone action and grossly underestimate the risk of endocrine disruption. This proposal will yield new knowledge about how such substances act in the body, or on wildlife, and form the basis for new more sensitive methods of environmental monitoring.Read moreRead less
