The investigation of protein folding – one the most important unsolved biological problems, is generating large amounts of complex data. This proposal aims to develop a highly collaborative content management system and interactive data mining tools that will allow the open exchange and analysis of protein folding data. A chief goal of this proposal is, by creating a rich informatics environment, to foster the exchange of ideas and rapid dissemination of new data across the international array of groups working in parallel on various aspects of the folding problem.

New Methods for Structural Biology in Solution. This project aims to expand the range of applications of nuclear magnetic resonance (NMR) spectroscopy in pharmaceutical research, where NMR spectroscopy is already used routinely for the identification of chemical compounds that bind to protein targets. The techniques developed aim at providing rapid and broadly applicable tools for 3D structure determinations of chemical compounds bound to their protein target, identification of protein-protein interaction sites and characterization of protein motions. The limits of NMR spectroscopy will be pushed to analyse systems of significantly increased molecular weights. The project includes applications to drug targets such as the dengue virus NS2B/NS3 protease.

New Methods for Structural Biology in Solution. New technologies will be developed that are sufficiently rapid and inexpensive to compete with and replace the mutagenesis experiments that biologists usually perform to identify and characterize the functionally important parts of a protein. Nuclear magnetic resonance (NMR) spectroscopy techniques in combination with various selective labelling schemes will be developed with the goal of identification and structural characterization of protein-ligand interactions at increased rates and enhanced accuracy. In addition, the three-dimensional structures of proteins and protein domains of biologically important functions and unknown fold will be determined by NMR. The project aims at techniques of direct impact in pharmaceutical industry.

Membrane Protein Structure and Interaction Facility. While it is estimated that a third of the human genome encodes for membrane proteins, the structures of only relatively few membrane proteins are currently known. It will be some time before membrane protein structure determination becomes routine, yet over 50% of the drugs on the market today rely on the activity of membrane proteins for their efficacy. This application seeks to establish a Membrane Protein Structure and Interaction Facility for the development and application of novel techniques and approaches to study the structure and interactions of membrane proteins. Research progress will be greatly enhanced by the establishment of this dedicated facility with cutting-edge technologies for the study of membrane proteins.

Mechanosensitive properties and modulation of N-methyl-D-aspartate (NMDA) receptors by lipid environment. This project will provide new information about the molecular determinants which influence NMDA receptor channel gating that will significantly advance our understanding of a link between NMDA receptor function and many neurodegenerative diseases as well as pain and learning and memory. The outcomes of this project may lead to the discovery of novel lipid-based biomaterials for application in medicine and the drug industry. This research is highly significant in relation to human health. The biological and nutritional aspects of polyunsaturated lipids and dietary fish oils have long been recognized. Thus this project will provide further knowledge that could benefit the health of the nation with consequent reduced health care costs.
