Neurotrophic factors for pelvic autonomic neurons: the role of neurturin. This project is about pelvic parasympathetic neurons, which are responsible for involuntary activities such as bladder voiding and penile erection. We are interested in the neurotrophic factors that determine survival of these neurons during early mammalian development and keep them healthy in adults. Little is known about ?parasympathetic neurotrophic factors? in general. This project is about pelvic parasympathetic neurons, which are responsible for involuntary activities such as bladder voiding and penile erection. We are interested in the neurotrophic factors that determine survival of these neurons during early mammalian development and keep them healthy in adults. Little is known about ?parasympathetic neurotrophic factors? in general. However we have recently discovered that the protein neurturin is very important in the pelvic parasympathetic system. We will determine exactly how neurturin affects pelvic neurons and how it interacts with other neurotrophic factors. Our results will fill a major gap in our knowledge of fundamental neurobiology.Read moreRead less
Genetics and evolution of Shigella O antigens. We use genome scale sequencing techniques to sequence 26 O-antigen gene clusters from Shigella. With the seven already known, this will give sequences for every O-antigen of Shigella. This will be the first time that such set is fully sequenced. Shigella are human specific pathogens, have emerged with the evolution of humans. O-antigens are important for their life and pathogenicity. O-antigens are important for their life and pathogenicity. This project will greatly extend our knowledge of the genetic basis and evolution of this important polymorphism. O-antigens are used for typing Shigella and also elicit strong immunity. The molecular data will help establish DNA based typing and vaccine development.Read moreRead less
Sociogenomics of honeybees. From genes to society. This project will show how complex social behaviour of honeybees like dance communication and thermoregulation is genetically controlled. The complete sequence of the honeybee genome will be published in 2003, and a gene chip based on it will be created shortly afterwards. These new technologies will make our project technically feasible. Age-matched workers that perform a behaviour like thermoregulation can be compared to sisters that do not, and the genes that are switched on in the two groups compared.Read moreRead less
Proteomic analysis of subcellular changes during apoptosis. This project aims to use a novel proteomic approach to examine mechanisms of apoptosis at the level of the plasma membrane, mitochondrion, nucleus and cytosol, screening protein extracts of cell organelles by the new technique of SELDI-TOF mass spectrometry in which proteins are adsorbed onto activated chips. This will provide protein mass profiles characteristic of various stages of apoptosis, and will allow identification of proteins ....Proteomic analysis of subcellular changes during apoptosis. This will provide protein mass profiles characteristic of various stages of apoptosis, and will allow identification of proteins of interest by conventional proteomic methods. The establishment of SELDI-MS as a viable tool for cell proteomics would open new opportunities to understand a broad range of cellular functions at the level of protein expression.Read moreRead less

The primary benefits are contribution to Australia's knowledge base and raising the profile of functional genomics in Australia, with the research priority of Frontier Technologies for Building and Transforming Australian Industries and priority goals in Breakthrough Science and Frontier Technologies. This project focuses on important biological questions surrounding gene regulation and sex chromosome evolution. International attention has already resulted in genome characterization of Australian icons (wallaby, Tasmanian devil and platypus), more research on these, and other Australian animals, will further highlight the importance of Australian fauna and impact positively on our scientific profile.Read moreRead less
Biosynthesis of nonribosomal peptide toxins in cyanobacteria: A functional characterisation of microcystin synthetase. Microcystins are potent toxins and tumour promoters produced by cyanobacteria associated with blue-green algal blooms. This non-ribosomal peptide is produced by microcystin synthetase, a unique enzyme complex comprised of peptide synthetases, polyketide synthases, and integrated accessory enzymes. We have identified and characterised the extensive gene cluster encoding this enzyme. This project describes the biochemical characterisation of specific enzyme activities within microcystin synthetase and how they determine the final structure and toxicity of the many forms of microcystin. Interactions between this enzyme complex and its substrate amino acids will provide information for the genetic engineering of this and similar natural products.Read moreRead less

The development of more generic and efficient Bayesian statistical methods will not only benefit applied statisticians but also the more occasional users of statistics in other disciplinary areas. The success of this project will enhance Australia's reputation as a strong contributor to the development of Bayesian methodologies. Two PhD students will also be provided training in computational Bayesian statistics.Read moreRead less
Stress, virulence and bacterial disease in temperate seaweeds: the rise of the microbes. Climate change is predicted to increase the spread and virulence of pathogens, and decrease the resistance to disease via temperature stress on the hosts. Combined with other human impacts (higher nutrients, pollution), we may be facing a major rise in the effect of disease on natural communities. However, these effects are largely unstudied. We will investigate the impact of marine pathogens on kelps and other seaweeds when they are stressed by temperature, elevated nutrients or other anthropogenic stressors. Kelp are the 'trees of the oceans', the organisms responsible for creating much of the habitat that fishes and other organisms live in. The loss of kelp forests due to disease would radically change these environments.Read moreRead less

My research will have important practical benefits for bioinformaticians and evolutionary biologists, because existing analytical methods will be rigorously tested and new tools will be developed. Australia has a comparatively high concentration of researchers in this field, so my research will foster domestic collaboration and import international expertise. The research will provide important insights into the rates and patterns of genetic changes associated with domestication, and into variation in evolutionary rates among the primate ancestors of humans. In addition to developing new software, which will be made publicly available, I will develop new evolutionary models to supplement existing software packages. Read moreRead less

Human analysis of huge quantities of relational data in biotechnology, web engineering, social networks, and computer networks requires visualization. In recent years, three-dimensional visualisation has promised to give new insights into such abstract data. However the past ten years of visualization research has had very little impact on the industry; visualization software providers have not adopted visualization methods developed by academics. We hypothesise that current 3D relational visualization methods fail because they do not pay attention to the human perception of topology. In this project we will leverage mathematical topology to construct new methods for the 3D visualization of relational data.
