Abstract: Barley (Hordeum vulgare L.) is one of the first domesticated grain crops and represents the fourth most important cereal source for human and animal consumption. BarleyVarDB is a database of barley genomic variation. It can be publicly accessible through the website at 146.118.64.11/BarleyVar. This database mainly provides three sets of information. First, there are 57 754 224 single nuclear polymorphisms (SNPs) and 3 600 663 insertions or deletions (InDels) included in BarleyVarDB, which were identified from high-coverage whole genome sequencing of 21 barley germplasm, including 8 wild barley accessions from 3 barley evolutionary original centers and 13 barley landraces from different continents. Second, it uses the latest barley genome reference and its annotation information publicly accessible, which has been achieved by the International Barley Genome Sequencing Consortium (IBSC). Third, 522 212 whole genome-wide microsatellites/simple sequence repeats (SSRs) were also included in this database, which were identified in the reference barley pseudo-molecular genome sequence. Additionally, several useful web-based applications are provided including JBrowse, BLAST and Primer3. Users can design PCR primers to asses polymorphic variants deposited in this database and use a user-friendly interface for accessing the barley reference genome. We envisage that the BarleyVarDB will benefit the barley genetic research community by providing access to all publicly available barley genomic variation information and barley reference genome as well as providing them with an ultra-high density of SNP and InDel markers for molecular breeding and identification of functional genes with important agronomic traits in barley. Abstract: In sexual assault cases and more specifically those involving childhood sexual abuse (CSA), victims may have had their potentially semen-stained clothing washed multiple times before a criminal investigation commences. Although it has been previously demonstrated that spermatozoa persist on cotton clothing following a single wash cycle, items of clothing washed multiple times are not routinely examined in these cases because of the assumption that the laundering process would have removed all seminal fluid and spermatozoa. The aim of this study was to examine the persistence of seminal fluid and spermatozoa on a range of fabric types including cotton, nylon, terry towel (100% cotton), polyester fleece, satin and lace which were laundered up to six times. Three techniques were used for the detection of seminal fluid and spermatozoa: an alternative light source, acid phosphatase test and microscopy. The study demonstrated that spermatozoa persisted on cotton and terry towel following six wash cycles. This data emphasises the need to recover and examine items of clothing and bedding of victims for semen, even if the item has been washed multiple times.

Abstract: The recently extinct New Zealand adzebills (Aptornithidae, Aptornis spp.) were an enigmatic group of large flightless birds that have long eluded precise taxonomic assignment as they do not closely resemble any extant birds. Adzebills were nearly wingless, weighed approximately 16–19 kg, and possessed massive adze-like reinforced bills whose function remains unknown. Using hybridisation enrichment and high-throughput sequencing of DNA extracted from subfossil bone and eggshell, near-complete mitochondrial genomes were successfully assembled from the two Quaternary adzebill species: the North Island Adzebill (Aptornis otidiformis) and South Island Adzebill (A. defossor). Molecular phylogenetic analyses confirm that adzebills are members of the Ralloidea (rails and allies) and are sister-taxon to the Sarothruridae, which our results suggest comprises the Madagascan wood rails (Mentocrex, two likely sp.) in addition to the tiny ( gram) rail-like Afro-Madagascan flufftails (Sarothrura, 9 spp.). Node age estimates indicate that the split between adzebills and Sarothruridae occurred ~39.6 Ma, suggesting that the ancestors of the adzebills arrived in New Zealand by long-distance dispersal rather than continental vicariance. This newly identified biogeographic link between physically distant New Zealand and Afro-Madagascar, echoed by the relationship between the New Zealand kiwi (Apterygiformes) and Madagascan elephant-birds (Aepyornithiformes), suggests that the adzebill’s near relatives were formerly more widespread. In addition, our estimate for the ergence time between the two Quaternary adzebill species (0.2–2.3 Ma) coincides with the emergence of a land-bridge between the North and South islands of New Zealand (ca. 1.5–2 Ma). This relatively recent ergence suggests that North Island adzebills are the result of a relatively recent dispersal from the South Island, from which the earliest (Miocene) adzebill fossil has been described.

Abstract: Cold case review teams and the processes that they adopt in their endeavour to solve historic crimes are varied and largely underreported. Of the limited literature surrounding the topic of cold case reviews, the focus is on clearance rates and the selection of cases for review. While multiple reports and reviews have been undertaken and recommend that the interface between investigators and forensic scientists be improved, there is little evidence of cold case teams comprised of a mixture of investigators and scientists or experts. With the growing reliance on forensic science as an aide to solvability, the authors propose that the inclusion of forensic scientists to the central cold case investigation may be a critical factor in future success. The paper aims to discuss this issue. To support the proposed approach, the authors conducted a review of the current literature seeking insight into the reported make-up of cold case teams. In conjunction with this, the authors reviewed a number of commissioned reports intended to improve cold case reviews and forensic services. While many of the reviewed reports and recommendations suggested better integration with scientists and external expertise, little evidence of this in practice was reported within published literature. Open dialogue and cross pollination between police investigators and forensic scientists are likely to mitigate biases, inform case file triage and better equip investigations with contemporary and cutting-edge scientific solutions to the evidence analysis for cold cases. Furthermore, with respect to scientists within academia, large pools of resources by way of student interns or researchers may be available to assist resource-sparse policing jurisdictions. To the authors’ knowledge, this is the first peer-reviewed recommendation for the consideration of integrated forensic scientists within a cold case review team. Multiple reports suggest the need for closer ties, but it is the anecdotal experience of the authors that the benefits of a blended task force approach may yield greater success.

Abstract: In forensic laboratories, increased extraction efficiency of trace evidence is paramount because analytical success is intrinsically dependent on the quantity of DNA recovered. Moreover, highly concentrated nucleic acids are vital for effective downstream analysis and high quality results. This study investigated the efficiency of extraction with the Qiagen® QIA ® DNA Investigator kit, and explored improvements to the methodology that would maximise the recovery of low concentration forensic s les. Controlled amounts of starting cellular material were used to mimic trace (or low level) DNA deposits prior to DNA extraction with the Investigator kit. Addition of the provided carrier RNA along with conducting two successive elutions of 50 µL improved the net recovery of DNA to 95%. Concentration with centrifugal filters post-extraction were able to concentrate DNA but a large net loss was observed. For the concentration of historic, retrospectively extracted DNA, centrifugal methods are able to concentrate DNA extracts previously too dilute for analysis. These concentrated volumes, however are small, allowing for minimal downstream analysis attempts before the s le is exhausted.

Abstract: Many factors affect the solvability of homicides, including body disposal location and time between death and recovery. The aim of this exploratory study was to probe a number of spatiotemporal variables for trends across a subset of solved homicide case data from 54 North American serial killers, active between 1920 and 2016 (125 solved cases) to identify areas for further research. We investigated murder site and body disposal site as location variables with eight subcategories across eight discrete time series, seeking insight into how these factors may affect the early stages of an investigation and (therefore by inference) solvability. The findings showed that bodies recovered after 48 hours are more likely discovered outdoor while those discovered within 24 hours, within the victim’s residence. This has implications for the ability to recover forensic evidence when bodes are located after a prolonged time since death as well as in more hostile environments.

Abstract: We report the preliminary observations of the peptide content of decomposition fluid produced under controlled laboratory conditions and in the absence of a soil matrix. Four domestic pig (Sus scrofa domesticus) cadavers were used to model human decomposition over a four-week trial period physical characteristics were recorded and the peptide components of decomposition fluid was analysed using high performance liquid chromatography-time of flight mass spectrometry. Preliminary data analysis indicated that a range of peptides were consistently detected across the course of the trial period and 27 of these were common to all four cadavers 22 originating from haemoglobin. The peptides associated with haemoglobin subunit alpha and beta displayed a breakdown pattern that remained consistent for all cadavers for the duration of the trial. Though identification of peptides during decomposition has potential for estimating the time since death, quantification of selected peptides is likely to be essential to identify time-dependent trends.

Abstract: A review was undertaken of the developments made with integrating forensic evidence into the analytical process to support police investigations. Evidence such as DNA, fingerprints, fibers, accelerants, tyre marks, and so forth, can support to differing degrees the various working theories or hypotheses about the nature of the alleged crime, the persons of interest and the modus operandi. Investigators however, either forensic or detective, bring various biases to evidence capture and analysis, biases which are better understood in the intelligence community. Structured analytical techniques have a long history in intelligence analysis, for ex le analysis of competing hypotheses, which serves several purposes: information sharing, clarity of communication, and to highlight the common forms of bias brought to bear in an investigation. We illustrate the representation of links based on traces and intelligence, and how these can be stored in databases permitting better “reasoning” with evidence. We also present some recommendations for integration of forensic intelligence into the investigative analytic process and review information systems in this area.
